Sample preparation To 1.0 g of the powdered herbal drug add 10 mL of methanol and heat in a water-bath at 65 °C under a reflux condenser for 5 min. Cool and filter.
Mobile phase Anhydrous formic acid, water, methyl ethyl ketone, ethyl acetate 10:10:30:50 (V/V)
Derivatization Heat the plate at 100-105°C. Spray the still-warm plate with a 10 g/L solution of diphenylboric acid aminoethyl ester in methanol, then spray with a 50 g/L solution of macrogol 400 in methanol; allow to dry in air for about 30 min and examine in ultraviolet light at 365 nm.
System Suitability Test (SST) Under development
Specific marker(s) Under shortwave UV (254 nm), prior to derivatization
  • chlorogenic acid: quenching zone at RF ~ 0.43
  • hyperoside: quenching zone at RF ~ 0.49
