Sample preparation Carefully crush 0.1 g of the herbal drug with a glass rod and moisten with 0.2 mL of water. After 3 min add 5 mL of methanol, allow to stand for 20 min, protected from light, and filter through a plug of glass wool.
Mobile phase Water, 2-propanol, ethyl acetate 10:25:65 (v/v/v)
Derivatization Treat with anisaldehyde solution (0.5 mL of anisaldehyde, 10 mL of glacial acetic acid, 85 mL of methanol and 5 mL of sulfuric acid.) and examine in daylight while heating at 100-105 °C for 5-10 min
