Sample preparation | Weigh 1.0 g of the freshly powdered sample and place it in a 50-mL conical flask, then add 25 mL of petroleum ether (60-80ºC) . Sonicate (220 W) the mixture for 1 h. Filter the mixture and wash the residue with 10 mL of petroleum ether. Discard the filtrate. Dry the residue in air. Transfer the dried residue to a 25-mL conical flask and add 10 mL of methanol. Sonicate (220 W) the mixture for 1 h. Filter and transfer the filtrate to a 10-mL volumetric flask. Make up to the mark with methanol. |
Mobile phase | Ethyl acetate, methanol, water 40:15:6 (v/v/v) |
Derivatization | Spray with anisaldehyde reagent and heat at 105°C until the spots or bands become visible |
Sample preparation | To 1 g of the powder, add 50 mL of petroleum ether (30-60°C) , heat and reflux for 1h, filter and discard the petroleum ether solution. Wash the residue with 25 mL petroleum ether, discard the petroleum ether solution. Evaporate the residue to dryness, add 30 mL of methanol, heat and reflux for 1h, filter and use the filtrate as the test solution |
Mobile phase | Chloroform, ethyl acetate, methanol, water 15:40:22:10 (v/v/v/v) |
Derivatization | Spray with phosphomolybdic acid reagent (dissolve 2 g of phosphomolybdic acid in 20 mL of water, add gently 30 mL of sulfuric acid, mix well), and heat at 105 °C until the spots appear |
Sample preparation | Add 1.0 g of powdered sample to 10 mL of methanol, heat under reflux for 10 min, cool, filter and use the filtrate |
Mobile phase | Ethyl acetate, methanol, water 7:3:1 (v/v/v) |
Derivatization | Spray with 10% sulfuric acid in ethanol, heat at 105°C until the spots appear |
